Using 5` Race to Amplify and Sequence Cdna Encoding Kinase1a Type Resistance Gene Family in Creeping Bentgrass.

The objective of this study was to target and amplify cDNA encoding kinase 1a type R gene sequences in creeping bentgrass. In order to gain further knowledge on nucleotide and aminoacid sequences, 5` RACE PCR were used. Amplified fragments of expected size using degenerate primers were isolated and cloned. Sequence analysis of clones and analysis of probable translation products revealed that all encoded amino acid sequences were highly conserved in creeping bentgras used in this study. Employing 5` RACE is very useful to identify upstream sequence information and cloning full-length cDNA encoding resistance genes in this species.
